Uncontrolled secretion of extracellular matrix (ECM) proteins such as collagen can lead to excessive scarring and fibrosis and compromise tissue function. Despite the widespread occurrence of fibrotic diseases and scarring, effective therapies are lacking.This invention relates to inhibitors of TANGO1 activity and methods of use thereof. In particular, the invention relates to peptide inhibitors capable of preventing TANGO1 heterodimerisation and/or homodimerisation and therby prevent collagen export from the endoplasmic reticulum. The inhibitors of the invention are particularly useful for use in methods of treating fibrotic diseases, such as scleroderma.
