Catalase-powered nanobots for crossing mucus barriers

PCT/EP2024/081138Oral and intravenous drug administration are widely used routes in disease therapy, particularly in oncology, aiming to distribute active compounds throughout the bloodstream to reach the intended target site. Nevertheless, therapeutic drugs often face challenges in terms of limited bioavailability and significant offtarget effects.The present invention belongs to the field of nanotechnology. In particular, the invention relates to enzyme powered nanobots useful in drug delivery. The nanobots of the invention are particularly useful for delivering drugs across mucus barriers. The present invention provides a complex for use in delivering a cargo cross a mucus barrier, the complex comprising a particle with a surface coated with catalase; and the cargo, wherein the catalase is attached over the whole surface of the particle. The invention also provides a pharmaceutical composition comprising the complex, and the complex or the pharmaceutical composition for use in a therapeutic, prophylactic, and/or diagnostic method.
